چکیده

OBJECTIVE To study the effects of deltamethrin on tyrosine hydroxylase in nigrostriatum of male rats. METHODS Sprague-Dawley rats were daily treated with deltamethrin at 6.25 or 12.5 mg/kg body weight by gavage for 10 days. Then HPLC-fluorescence detection was used to analyze the contents of dopamine (DA), 3,4-dihydroxyphenylacetic acid (DOPAC) and homoranillic acid (HVA) in substantial nigra and striatum. The activities of tyrosine hydroxylase (TH) were also detected by HPLC-fluorescence detection. TH mRNA or TH protein levels were measured by RT-PCR and immunohistochemistry method. RESULTS The content of DA in striatum was significantly decreased by the treatments, suggesting an inhibition of DA synthesis by deltamethrin. The contents of DA metabolites DOPAC and HVA increased, indicating increased dopamine turnover. Furthermore, deltamethrin significantly decreased the activity, as well as the mRNA and protein levels of TH. CONCLUSIONS These findings reveal a novel aspect of deltamethrin neurotoxicity and suggest tyrosine hydroxylase as a molecular target of deltamethin on dopamine metabolism in the nigrostriatal pathway.

Reconstruction of the nigrostriatal pathway by simultaneous intrastriatal and intranigral dopaminergic transplants.

The main strategy in experimental and clinical neural transplantation in Parkinson's disease has been to place fetal nigral grafts not in their ontogenic site (substantia nigra) but in their target area (striatum). The reason for this ectopic placement is the apparent inability of nigral grafts placed in the ventral mesencephalon (VM) of the adult host to grow axons for long distances that are ...

Investigations into Potential Extrasynaptic Communication between the Dopaminergic and Nitrergic Systems

Nitric oxide is unconstrained by cell membranes and can therefore act along a broad distance as a volume transmitter. Spillover of nitric oxide between neurons may have a major impact on central nervous system diseases and particularly on neurodegeneration. There is evidence whereby communication between nitrergic and dopaminergic systems plays an essential role in the control of the nigrostria...

Testosterone Induces Molecular Changes in Dopamine Signaling Pathway Molecules in the Adolescent Male Rat Nigrostriatal Pathway

Adolescent males have an increased risk of developing schizophrenia, implicating testosterone in the precipitation of dopamine-related psychopathology. Evidence from adult rodent brain indicates that testosterone can modulate nigrostriatal dopamine. However, studies are required to understand the role testosterone plays in maturation of dopamine pathways during adolescence and to elucidate the ...

Influence of Striatal Astrocyte Dysfunction on Locomotor Activity in Dopamine-Depleted Rats

Introduction: Astrocyte dysfunction is the common pathology resulting in failure of astrocyte-neuron interaction in neurological diseases, including Parkinson’s Disease (PD). To date, only few experimental models of selective ablation of astrocytes are known. The aim of present study was to evaluate the effect of striatal injections of selective glial toxin L-aminoadipic acid (L-AA) on the loco...

Ultrastructural relations between nigrostriatal dopaminergic neurons and cholinergic nerve endings in the human brain.

The connections between cholinergic nerve endings and nigrostriatal dopaminergic neurons were studied in the substantia nigra pars compacta of the human brain. Immunocytochemistry of tyrosine hydroxylase followed by ionic fixation of acetylcholine-like cation in synaptic vesicles allowed dopaminergic neurons and cholinergic nerve endings to be visualized on the same ultrathin section. Numerous ...
